Inhibitory effects of connective tissue growth factor siRNA in high glucose induced connective tissue growth factor and collagen synthesis in rat mesangial cells

Abstract: Objective To investigate the inhibitory effects of siRNA targeting CTGF on the expression of CTGF and collagen type Ⅰ in cultured rat mesangial cell (RMC) with high glucose. Methods SiRNA targeting CTGF was designed, synthesized and transfected into RMC cultured with high glucose. Expression of CTGF and col-lagen type Ⅰ was examined by reverse transcription polymerase chain reaction(RT-PCR) and western blot. Results After siRNA was transfected, expression of CTGF and collagen type Ⅰ decreased significantly compared with that in RMC cultured with only high glucose. Conclusion The siRNA targeting CTGF could inhibit synthesis of CTGF and collagen type Ⅰ in RMC cultured with high glucose.
