Abstract: Objective To find an appropriate way to establish the mice model of chronic thromboembolism pulmonary hypertension (CTEPH) with simple manipulation,high success and feasibility,low cost and similar to human pathophysiology.Methods C57BL/6J mice were randomly divided into normal group (n =14),sham group (n =11),model group (n =22),and tranexamic acid group (n =9).Acute pulmonary thromboembolism was established by injecting autologous blood thrombi to jugular vein once a week,totally four weeks.At appointed time of the beginning of the sixth week,right ventricular systolic pressure (RVSP) of mice was measured via direct cardiac puncture using a closed chest technique and the heart and lung tissues were collected.The right ventricular hypertrophy index (RVHI) was measured.The established models were evaluated by morphological observation of lung tissue,histological section of hematoxylin-eosin staining to evidence thrombi in tissues.Results RVSP were elevated in model group compared to normal group and sham group [(19.96± 0.57) mmHg vs (17.18±1.17) mmHg,(16.73±0.61) mmHg,P <0.05 orP<0.01].RVHI in model group was higher than that in normal group,sham group and tranexamic acid group (29.60 % ± 1.69 % vs 19.55 % ± 1.16%,19.80± 1.09%,18.07± 0.61%,all P <0.001).No obvious thrombi were detected in the histological section of lung tissues of model group mice at the sixth week.However,compared with sham group,it was found that medial smooth muscle layer was thicken,the cavity became narrow or nearly closed and inflammatory cell infiltration in pulmonary arterioles of the model group.Conclusions The method to set up CTEPH mice model may give some hints of this disorder from the current data.
