Abstract: Objective To detect the expression of IL-23 receptor on spleen CD4+ T lymphocytes in mouse model of acute bronchial asthma (asthma)as well as its significance in the pathogenesis. Methods Spleen CD4+ T lymphocytes origin from spleen of mouse model of acute asthma isolated by immunomagnetic beads and they were used to determine the expression of IL-23 receptor mRNA by realtime PCR,the level of IL-23 receptor protein by Western blotting,the ratio of positive Th17 cells by flow cytometer and the production of IL-17 by ELISA. Results The expression of IL-23 receptor,the ratio of Thl7 cells and the production of IL-17 on Spleen CD4+ T lymphocytes in asthmatic model was obviously higher than that of the control group. The expression of IL-23 receptor on spleen CD4+ T lymphocytes positively correlated with the ratio of positive Th17 cells and the production of IL-17. Conclusions IL-23receptor possibly plays an important role in the pathogenesis of asthma.
