Abstract: Objective To establish an HPLC method for simultaneous determination of three components(ellagic acid、gallic acid and rutin)in bran-processed rosae laevigatae fructus.Methods Determination by HPLC,chromatographic column:Cosmosil 5C18-MS-Ⅱcolumn(250 mm×4.6 mm,5 μm);mobile phase:acetonitrile(A)-0.05%phosphoric acid aqueous solution(B),gradient elution;the flow rate was 1.0 mL/min;column temperature was 30℃;detection wavelength was 272 nm;injection volume was 10 μL.Results The standard cures of gallic acid,rutin,and ellagic acid good linearity in 9.195 8-45.978 8 μg/mL,19.382 6-193.826 0 μg/mL,10.923 0-54.615 0 μg/mL(r=0.999 8,0.999 2,0.999 9,respectively);the average sample recovery rates(n=6)were 93.25%、92.18%and 92.95%,respectively,with RSDs of 1.2%、1.7%and 1.9%.Conclusion This study can provide a reference for the determination standard of flavonoids in bran-processed rosae laevigatae fructus.The established method for the determination of gallic acid,rutin,and ellagic acid has good repeatability,stability,and reliability,and can provide reference for the quality control of bran-processed rosae laevigatae fructus.
