Expression of PTEN, Lgr5 and β-catenin in intestinal injury and repair model

Abstract: Objective To establish necrotizing enterocolitis (NEC) model as non-operative NEC patients in neonatal rat,and observe the expression variety of PTEN,Lgr5 and β-catenin at different time after NEC,aiming to explore the role of PTEN,Lgr5 and β-catenin in intestinal ischemic injury and biological repair.Methods The hypoxia and cold stimulation were conducted to establish intestine repair NEC model in neonatal rats.Neonatal rats were divided into control and experimental groups.Ileocecal bowel tissues were harvested different time points (24,72 and 120 h) after last hypoxia and cold administration.The PTEN,Lgr5 and β-catenin mRNA expression levels in intestinal tissue was detected via real-time PCR.Results NEC model was successfully established through administration of hypoxia and cold.Compared the control group,PTEN,Lgr5 and β-catenin mRNA expression levels in experimental group were significantly higher at 24 h (P<0.05).However,their expression increasing was not significantly at 72 h and 120 h.Conclusions Hypoxia and cold can be used to establish NEC model.In early phase of intestine injury,PTEN,Lgr5 and β-catenin mRNA expression levers increased,which probably correlates to promotion of stem cells in intestinal tissue.While in late phase of intestine injury and repair,their expression increasing is not significantly,which is the reflect to negative regulative activity of β-catenin and stem cells proliferation speed,as well as stem number.
