Abstract: Aim To study the correlation between NLRP3 inflammasome,inflammatory response,and pyroptosis markers in patients with chronic periodontitis and Porphyromonas gingivalis infection.Methods 300 patients with chronic periodontitis were selected and divided into a positive group and a negative group based on the infection status of Porphyromonas gingivalis.The expres-sion levels of NLRP3,apoptosis-associated speck-like protein(ASC),Caspase-1 mRNA and GSDMD-N protein in periodontal tissue,as well as interleukin-1β(IL-1β),IL-8,tumor necrosis factor-α(TNF-α)in gingival crevicular fluid between two groups were meas-ured.Correlation analysis was done by using Pearson test.Results Plaque index(PLI),gingival index(GI),attachment loss(AL),pocket depth(PD),periodontal tissue NLRP3,ASC,Caspase-1 mRNA,GSDMD-N protein,and gingival crevicular fluid IL-1β、IL-18,TNF-α levels in positive groups were higher than those in the negative group(P<0.05).The expression levels of NL-RP3,ASC,and Caspase-1 mRNA were positively correlated to PLI,GI,AL,PD,GSDMD-N proteins,IL-1 β,IL-18,and TNF-αlevels(P<0.05).Conclusion The infection of Porphyromonas gingivalis in patients with chronic periodontitis exacerbates the condition,inflammatory response,and necrosis,and its effect may be related with the activation of NLRP3 inflammasomes.
