Effect of "Biao-Ben points association" electro-acupuncture combined with dietary restriction on SIRT1 and UCP1 expression in brown adipose tissue of obese rats

Abstract: 目的:研究“标本配穴”电针和饮食控制对肥胖大鼠褐色脂肪组织沉默信息调节因子1(silent information regulator1,SIRT1)和解偶联蛋白(uncoupling protein,UCP1)的影响,比较其单独及协同作用.方法:将40只高脂喂养的肥胖大鼠随机分为4组,每组10只.模型对照组(C)继续高脂喂养,电针组(E)给予“标本配穴”电针治疗,限食组(R)每只给予对照组摄食量的70%,电针+限食组(E+R)给予“标本配穴”电针结合限食干预治疗,共电针及限食8 wk.检测各组大鼠体质量和血清葡萄糖(fasting plasma glucose,FPG)、胰岛素(fasting insulin,FINS)、甘油三酯(triglyeride,TG)、总胆固醇(total cholesterol,TC)水平变化,对褐色脂肪组织进行SIRT1蛋白和UCP1mRNA的测定.结果:与C组比较,3个电针和饮食干预组体质量及FPG、FINS、TG和TC均明显下降(P<0.01),且E+R组下降程度优于单独电针或限食组P<0.05);与C组比较,3个干预组褐脂中SIRT1蛋白表达水平明显升高(P<0.01);与C组比较,3个干预组褐脂中UCPl mRNA表达明显升高(P<0.01),且E+R组效果明显优于单独电针或单独饮食干预组(P<0.05).结论:“标本配穴”电针和限食可能是通过促进褐色脂肪组织中SIRT1和UCP1的表达,增加产热而发挥减肥作用.“标本配穴”电针法抵抗肥胖的功效与饮食控制相当,二者之间存在一定的协同作用.
