Abstract: Objective To assess the value of ADAM 12(a disintegrin and metalloprotease 12) levels in second-trimester maternal serum as a potential marker for preeclampsia. Methods Using preliminarily developed indirect competitive ELISA,a casc-control study was performed.Serum samples were collected every week from 14 to 18 weeks of gestation from 100 women who later developed preeclampsia and 200 normal gravidas.Serum ADAM 12 levels were assayed and converted to MoM according to different gestational age and compared. Results The median levels of serum ADAM 12 from 14 to 18 weeks of gestation in normal pregnant women were 680μg/L,738μg/L,801μg/L,849μg/L and 900 μg/L respectively, and 598μg/L,664 μg/L,729μg/L,791μg/L and 839μg/L in preeclamptic women. The serum ADAM 12 levels in preeclamptic women had a mean log MoM of-0.05242,which was significantly lower than that of the normal groups(-0.00368,P<0.01).Conclusions The maternal serum levels of ADAM 12 are significantly lower in women who later developed preeclampsia from 14 to 18 weeks of gestation compared with normal pregnancies. Thus, second trimester serum ADAM 12 may be served as a marker for preeclampsia screening.
