The effects of hyperbaric oxygen therapy of hypoxicischemic brain injury on levels of neuropeptide Y136 and calcitonin gene-related peptide in neonatal rats

Abstract: 目的观察高压氧(HBO)治疗新生大鼠缺氧缺血性脑损伤后血浆和大脑皮层的神经肽Y1-36(NPY)和降钙素基因相关肽(CGRP)含量的动态变化.方法 7 d龄新生大鼠左侧颈总动脉结扎形成缺血,并吸低氧氮混合气形成缺氧,随机分为脑缺氧缺血+常氧高压氮组、脑缺氧缺血+HBO治疗组、假手术组和正常对照组,再按不同时间各分成4个小组.新生大鼠缺氧缺血性脑损伤后即刻行HBO治疗,用放射免疫分析法测定治疗后不同时间血浆和大脑皮层的神经肽Y1-36和降钙素基因相关肽免疫活性物质(ir-NPY、ir-CGRP)含量的变化.结果缺氧缺血性脑损伤后不同时间,血浆ir-NPY和ir-CGRP、双侧皮层ir-NPY、伤侧皮层ir-CGRP含量不同程度升高;HBO治疗,可部分抑制血浆ir-NPY、ir-CGRP的升高,左侧皮层ir-NPY水平的升高被明显抑制,右侧皮层ir-CGRP水平受影响较小.结论缺氧缺血性脑损伤的发生发展与外周及中枢神经肽Y1-36和降钙素基因相关肽的过量生成有关.HBO治疗降低外周神经肽Y1-36和降钙素基因相关肽,降低伤侧脑区神经肽Y1-36的水平,可能是减轻缺氧缺血性脑损伤的作用途径之一.
