Comparison of the expression of GRP78 in omental fat between normal weight and obese women patients

Abstract： 目的 探讨葡萄糖调节蛋白78(GRP78)在肥胖及肥胖相关疾病中的作用. 方法 将30例40～60岁女性患者分为3组:正常对照组(NC组,BMI＜23 kg/m2),超重组(OW组,BMI＜28 kg/m2)和肥胖组(OB组,BMI＞28 kg/m2),每组10例.在其腹部择期手术时取其网膜脂肪组织,应用RT-PCR 和Western-blot检测上述三组患者脂肪组织中GRP78的表达水平. 结果 OB组GRP78 mRNA与蛋白水平(GRP78/β-actin)显著高于对照组(P＜0.05).GRP78/β-actin与BMI呈正相关(r≥0.75,P＜0.05). 结论 GRP78表达量与BMI呈正相关(GRP78蛋白及mRNA水平OB组＞OW组＞NC组).GRP78可作为预测肥胖及T2DM和心血管疾病危险因素的指标之一.
