Abstract: Objective To explore the changes of homing related activities of cord blood (CB)hemato- poietic cells during ex vivo culture supported by human bone marrow stromal cells and cytokines. Methods Mononuclear cells(MNCs) separated from CB were cultured in a serum-free with only FBMSC or only exogenous cytokines support or with both of them support culture system. On day 0、10 and 14, CD34+ CXCR+ and CD34+ VLA-4+cells were quantitated by FACS. Results After culturing, CD34+CXCR4 + cells and CD34+ VLA-4+ cells were ignificantly increased. Compared with other groups, expansion fold of CD34+ CXCR4+/CD34+ VLA- 4+cells was higher in FBMSC and cytokines supported group(P < 0. 05). Conclusion The culture system used in the study can support the expansion of hematopoietic stem and progenitor cells which had homing related ca- pacity. This culture system may be a feasible way for in vitro culture of cord blood cells.
