SNRPD3 protein expression level in nasopharyngeal carcinoma and its correlation with radiosensitivity

Abstract: Objective:To investigate the correlation between the expression level of SNRPD3 protein in nasopharyngeal carcinoma (NPC) and radiosensitivity of NPC.Methods:The clinical data and paraffin-embedded NPC tissue samples of 133 patients with newly diagnosed non-metastatic NPC, who received radiotherapy in the Sun Yat-sen University Cancer Center between 2016 and 2018, were retrospectively included in the study. The expression of SNRPD3 protein in the NPC tissue samples was detected by immunohistochemistry, and the level of SNRPD3 protein expression was evaluated according to the staining index. The tissue samples were divided into high- and low-SNRPD3 expression groups. Kaplan-Meier method was used for survival analysis, Log-Rank test was used to compare the survival differences, and Cox model was used to analyze the relationship between the expression level of SNRPD3 protein and radiosensitivity of NPC.Results:The rate of SNRPD3 protein expression in NPC tissues was 97.7% (130/133) . The expression level was positively correlated with the recurrence ( P=0.036) and deaths ( P=0.023) of NPC patients after treatment. The proportion of patients with high expression of SNRPD3 protein in the radiosensitive group was significantly lower than that in the radioresistant group ( P=0.036) . The 5-year locoregional relapse-free survival (LRRFS) (75.9% vs. 91.4%, P=0.038) and overall survival (OS) (77.5% vs. 92.8%, P=0.042) in the high SNRPD3 expression group were lower than those in the low SNRPD3 expression group. The expression level of SNRPD3 protein was an independent prognostic factor for 5-year LRRFS ( P=0.041) and OS ( P=0.044) in NPC. Conclusion:The SNRPD3 protein is generally highly expressed in NPC tissues, and its expression level is negatively correlated with radiosensitivity of NPC. SNRPD3 is promising as a biomarker for predicting the radiosensitivity of NPC.
