Abstract: Objective To study the effect of down-regulation of ID-1 on invasion,migration and secretion of immunosuppressive factors in HuH-7 cells.Methods Realtime PCR and Western blot were used to detect the expression of ID-1 in human hepatocellular carcinoma HuH-7,MHCC97H,SNU-449 cells and normal hepatic L-02 cells.ID-1 siRNA was transfected into HuH-7 cells.Transwell chamber was used to measure invasion and migration.The levels of IL-6 and IL-1β secreted by cells were detected by ELISA.The levels of MMP-2,MMP-9 and NF-κBp65 were detected by Western blotting.HuH-7cells transfected with ID-1 siRNA were treated with NF-κB activator,cell invasion,migration and secretion of IL-6 and IL-1β were detected.Results The level of ID-1 in HuH-7,MHCC97H and SNU-449 cells was higher than that in normal liver L-02 cells,the ID-1 level in HuH-7 cells was the highest.The invasion and migration ability of HuH-7 cells transfected with ID-1 siRNA decreased,the secretion of IL-6 and IL-1β decreased,the levels of MMP-2,MMP-9 and NF-κBp65 in cells decreased,NF-kappa B activator reverses the effects of ID-1 siRNA on invasion,migration and secretion of IL-6 and IL-1β in HuH-7 cells.Conclusion Downregulation of ID-1 reduces the ability of HuH-7 cells to invade,migrate and secrete immunosuppressive factors by inhibiting activation of NF-κB signal.
