Effects of Resveratrol on expression of autophagy-related proteins in macrophages with oxidative injury

Abstract: Objective To investigate the effects of Resveratrol (Res) on expression of autophagy-related proteins in oxidatively damaged macrophages.Methods Phorbol ester was used to induce differentiation of human mononuclear THP-1 cells into macrophages.A model of macrophage with oxidative injury was established and used to determine the optimal concentration and duration of H2O2 induced oxidative injury.The macrophages were divided into control group,H2O2 group,Res+H2O2 group,and 3-methyladenine (3-MA)+Res+H2O2 group.Western blotting was used to detect the expression levels of LC3I,LC3 Ⅱ and Beclin-1 proteins.Results Compared with the control group,the viability of macrophages in the H2O2 group was significantly decreased.The ratio of LC3 Ⅱ to LC3 Ⅰ and the expression level of Beclin 1 protein in macrophages of H2O2 group were significantly decreased.Compared with the H2O2 group,the macrophage viability was significantly increased after treatment with different concentrations of Res (Res+ H2O2 group) [H2O2 group (50.17± 1.39) %;40 μmol/L Res group (91.03±4.70) %],and the ratio of LC3 Ⅱ to LC3 Ⅰ and Beclin 1 protein expression level were also significantly increased.Compared with Res+H2O2 group,the ratio of LC3 Ⅱ to LC3 Ⅰ and Beclin 1 protein expression level were significantly decreased in the 3-MA+Res+H2O2 group.Conclusion Resveratrol may ameliorate the oxidative damage in macrophages.The underlying mechanism may be up-regulation of autophagy-related proteins and thereby modulation of autophagy in certain cells.
