Astragaloside Ⅳ attenuates cerebral ischemia and reperfusion injury and reduces activation of NLRP3 inflammasome and NF-κB phosphorylation in rats following a transient middle cerebral artery occlusion

Abstract: 本文旨在探讨黄芪甲苷(astragaloside Ⅳ,AST-Ⅳ)在大鼠脑缺血再灌注损伤中的保护作用及其抗炎机制.采用改良线栓法制备大鼠大脑中动脉栓塞脑缺血再灌注模型,以神经功能缺失评分和脑梗死体积综合评价AST-Ⅳ抗脑缺血再灌注损伤的作用,Western blot检测脑组织中NLRP3、ASC、pro-Caspase-1、Caspase-1、pro-IL-1β、IL-1β、pro-IL-18、IL-18、磷酸化NF-κB和总NF-κB蛋白表达水平.结果 显示,与模型组比较,AST-Ⅳ能降低大鼠神经功能缺失评分,减少脑梗死体积,降低脑组织中NLRP3、Caspase-1、pro-IL-1β、IL-1β、pro-IL-18和IL-18蛋白水平,并抑制磷酸化NF-κB蛋白表达.以上结果提示,AST-Ⅳ具有抗脑缺血再灌注损伤作用,其机制可能与抑制NF-κB蛋白磷酸化以及抑制NLRP3炎症小体活化有关.
