Abstract: 本文旨在研究原癌基因c-src在体外培养的9日龄大鼠精原干细胞中的表达及其与精原干细胞活性相关的信号通路.用MTT比色法观察反义c-src脱氧寡核苷酸(oligodeoxynucleotides, ODNs)对精原干细胞作用的量效及时效关系;用RT-PCR检测c-src mRNA的表达;用Western blot检测c-src表达产物pp60c-src和磷酸化的信号转导与转录激活子-3(phosphorylated signal transducer and activator of transcription. 3, p-STAT3)的表达.结果显示,与空白对照组相比,10μmol/L反义c-src ODNs作用12h后精原干细胞活性下降8.1%(P<0.05),且c-src mRNA表达明显下调;Western blot结果显示反义c-src ODNs组pp60c-src、p-STAT3蛋白表达与空白对照组相比分别下降了33.8%、45.3%(均P<0.01).以上结果提示,原癌基因c-src可能通过p-STAT3蛋白影响精原干细胞的活性.
