MiR-637 inhibits cell migration and invasion and promotes apoptosis in gastric cancer cells by targeting ERBB3

Abstract: 背景研究发现miR-637可抑制结肠癌、甲状腺乳头状癌、胶质瘤等多种肿瘤细胞的侵袭、迁移;其在胃癌(gastric cancer,GC)中下调表达,但对GC细胞迁移、侵袭及凋亡的影响及分子机制尚不清楚.目的探讨miR-637对GC细胞迁移、侵袭及凋亡的影响及其作用机制.方法实验设置miR-NC组、miR-637组、anti-miR-NC组、anti-miR-637组、si-NC组、si-ERBB3组、miR-637+pcDNA3.1组、miR-637+pcDNA3.1-ERBB3组、miR-NC+WT-ERBB3组、miR-NC+MUT-ERBB3组、miR-637+WT-ERBB3组、miR-637+MUT-ERBB3组;qRT-PCR检测miR-637的表达水平;Western blot检测蛋白表达;Transwell检测细胞迁移、侵袭;流式细胞术检测细胞凋亡;双荧光素酶报告基因检测实验检测荧光活性.结果相较于癌旁组织,GC组织中miR-637的表达水平显著降低(P<0.05);过表达miR-637和抑制人类表皮生长因子受体3(human epidermal growth factor receptor 3,HER3/ERBB3)表达可抑制基质金属蛋白酶2和B细胞淋巴瘤/白血病-2(B cell lymphoma/leukemia-2,Bcl-2)的表达,促进上皮钙黏蛋白和Bcl-2相关X蛋白(Bcl-2 associated X protein,Bax)的表达;抑制GC细胞SGC-7901迁移、侵袭,促进细胞凋亡.miR-637靶向调控ERBB3的表达,过表达ERBB3能逆转miR-637对GC细胞SGC-7901迁移、侵袭抑制和凋亡促进的作用.结论miR-637可抑制GC细胞SGC-7901迁移、侵袭,促进细胞凋亡,其机制可能与靶向下调ERBB3的表达有关,将可为GC的预防和治疗提供新靶点.
