Efficacy of Shuanghu liver-clearing granule in prevention and treatment of carbon tetrachloride-induced hepatic fibrosis in rats

Abstract: 目的:探讨双虎清肝颗粒对四氯化碳诱发大鼠肝纤维化的防治作用.方法:清洁级SD大鼠60只,随机分成6组:A组(正常对照组)、B组(空白模型组)、C1组(双虎清肝颗粒小剂量组)、C2组(双虎清肝颗粒中剂量组)、C3组(双虎清肝颗粒大剂量组)、D组(水飞蓟素对照组).除空白对照组外,其余各组大鼠均经皮下注射400 mL/L四氯化碳8 wk,各治疗组再给予不同剂量双虎清肝颗粒干预8 wk,阳性对照组给予水飞蓟素干预8wk,化学发光法检测6组大鼠血清肝纤维化指标,取大鼠肝脏作组织病理学检查.结果:C3、D组同B组相比,脂质空泡明显减少,肝细胞坏死程度明显减轻,假小叶形成、胶原纤维沉积明显减少.C1、C2、C3和D组大鼠纤维化程度与B组比较,有差异(12.6±4.6,10.7±3.9,6.4±3.4,6.5±3.3 vs16.1±5.5,均P<0.05).C3、D组大鼠血清透明质酸酶(HA)、血清Ⅳ型胶原(ⅣC)及血清Ⅲ型前胶原(PⅢP)水平较B组降低(161.50±37.38,185.30±9.76 vs 292.25±72.99:18.55±2.27,17.60±2.54 vs 33.42±6.49:19.49±3.26,25.84±4.69vs 36.58±8.13,P<0.001或0.05).结论:双虎清肝颗粒能够显著改善四氯化碳所诱发的大鼠肝纤维化,并且量效关系明显.
