Preliminary study on effect of indomethacin and mibolerone on Hep-2 cell proliferation and apoptosis

Abstract： 目的 探索吲哚美辛(IND)与雄激素(Mib)对喉癌细胞增生与凋亡的作用.方法 Hep-2细胞暴露于含IND(125,250,500μmol/L),或Mib(1,10,100 nmol/L),或IND与维持剂量的Mib(1 nmol/L)同时存在的培养基中,分别培养24 h;用MTT法、锥虫蓝染色法与流式细胞仪分析法分别检测细胞增生、细胞活力、细胞周期和细胞凋亡.结果 IND使Hep-2细胞增生及细胞活力明显降低,S期细胞含量明显增加,且出现凋亡峰;Mib(100 nmol/L)明显促进Hep-2细胞增生;Mib(1 nmol/L)与IND(250μmol/L)同时作用,下调细胞增生的抑制率,明显减少死细胞数,但S期细胞含量以及细胞凋亡比率明显上升.结论 IND强烈抑制Hep-2细胞增生与细胞活力,阻滞细胞从S期向G2/M期转化,并诱导其凋亡;维持剂量的Mib具有保护细胞膜的作用,而另一方面又具有促进IND诱导细胞凋亡的作用.
