Effect of hypoxia-inducible factor-2α on VEGF expression in villous explants under hypoxia

Abstract: 目的探讨缺氧对体外培养的早孕期绒毛组织中血管内皮生长因子(vascular endothelial growth factor,VEGF)表达水平的影响,以及应用反义寡核苷酸降调缺氧诱导因子2α(hypoxia inducible factor-2α,HIF-2α)后,VEGF表达水平的变化.方法取孕6~8周的绒毛组织,分四组进行体外培养:常氧浓度组、低氧浓度组、HIF-2α正义组和HIF-2α反义组.将HIF-2α正义或反义寡核苷酸分别加入后两组的培养液中,进行缺氧培养.48 h后,收集绒毛组织,应用Western印迹方法,分别检测各组HIF-2α蛋白的表达水平;并应用荧光定量PCR检测HIF-2α和VEGF mRNA表达水平.结果与常氧浓度组相比,低氧浓度组的HIF-2α mRNA(54.46±4.27与29.64±1.63)和VEGFmRNA(143.53±2.82与6.47±1.63)表达及HIF-2α蛋白(62.16±1.35与32.04±2.14)表达均增加(P<0.05);与HIF-2α正义组相比,HIF-2α反义组的HIF-2α蛋白(33.68±2.24与63.80±1.58)表达水平降低,HIF-2α mRNA(24.50±2.90与55.60±1.73)和VEGF mRNA(5.38±1.60与144.91±2.48)表达水平也下降(P<0.05).结论缺氧可诱导孕早期绒毛组织VEGF的表达增加,这种作用可能是通过HIF-2介导的.
