Abstract: 目的探讨重组Endostatin基因(EScDNA)对大鼠胶质瘤的治疗效果及肿瘤微血管密度的变化.方法对质粒包埋的Endostatin基因(pSecTagA-EScDNA)测序鉴定后,转染人脐静脉内皮细胞(ECV-304),验证Endostatin基因对血管内皮细胞增殖的抑制作用;以C6胶质瘤细胞植于SD大鼠腋部皮下制作胶质瘤模型,将pSecTagA-EScDNA直接瘤内注射.分别观察记录肿瘤体积,绘制肿瘤生长曲线:对肿瘤标本进行微血管标记、计数,并比较治疗前后肿瘤微血管数量的变化.结果重组Endostatin基因治疗后的肿瘤生长明显受抑制,使肿瘤退缩至休眠甚至完全消失;肿瘤微血管密度明显低于未治疗者.结论重组Endostatin基因通过抑制肿瘤血管新生对大鼠胶质瘤有良好的治疗效果.
