An experimental research of neuroglobin expression changes after diffuse traumatic brain injury

Abstract: Objective To study the expression changes of neuroglobin in rats with the model of diffuse traumatic brain injury in order to explore the relationship between the neuroglobin and traumatic brain injury. Method The diffuse traumatic brain injury modal of rats was induced by the Marmarou' weight-drop' device. The quantitative real-time PCR and immunohistochemical techniques were used to detect the expression changes of neuroglobin in rat brain at different time points post-injury. Results The nucleic acid expression of neuroglobin reached its first peak at 0. 5 hour post-injury, and then it decreased gradually. At 6 hours post-injury,it recovered to the normal leveL At the time of 12 hours post-injury, the expression of neuroglobin increased again, and reached its second peak at 48 hours post-injury. After that time, it dropped down,but still maintained a higher level till 120 hours days post-injury. Similar to nucleic acid changes, the protein expression changes of neuroglobin had ' two-peak-changes' post-injury. At 2 hours and 72 hours post-injury, the expression of neuroglobin increased and reached its peak level respectively. Conclusions The expression of neuroglobin shows a characteristic of' two-peak-changes' after traumatic brain injury in rat brain. It suggested that neuroglobin may have a protective action to neuron suffering traumatic injury.
