The therapy of embryonic stem cell transplantation for bleomycin-induced pulmonary injury in mice

Abstract: Objective To investigate the curative effectiveness and action mechanism of embryonic stem cells(ESCs)transplantation for bleomycin-induced pulmonary injury in mice.Methods Sixty C57BL/6J female mice were divided randomly into six groups:control group,bleomycin model group,transplantation group 1,transplantation group 2,transplantation group 3 and transplantation group 4.Those mice except in control group were administrated bleomycin intratraeheally to induce lung injury model,then epithelial progenitor cells from S8-ESC were transplanted into mice from one time to four times respectively in four treatment groups,and the mice in control group and bleomycin model group were administrated 0.9%sodium chloride respectively.After 28 days,the life-span,hydrocyproline concentration in lung tissue,transforming growth factor β_1(TGF-β_1)and interferon-γ (IFN-γ)concentration in lung tissue homogenate were examined,and the pathologic changes of the lung and the engraftment of the epithelial progenitor cells in the injured lung were observed.Results (1)After 28 days,the death rate was 0%(control group),60%(bleomycin model group),50%(transplantation group 1),40%(transplantation group 2),40%(transplantation group 3)and 20%(transplantation group 4)respectively. (2)With the prolonged time of transplanting epithelial progenitor cells to the mice,hydrocyproline and TGF-β_1 concentrations in lung tissue homogenate in bleomycin model group,transplantation group 1,transplantation group 2,transplantation group 3 andtransplantation group 4 became lower and lower,while those in control group were normal.However,an inverse tendency was found in IFN-γ concentrations.Comparisons between each group had significant difference(P<0.05).(3)With the prolonged time of transplanting epithelial progenitor cells to the mice,the severity of the lung injury in bleomycin model group,transplantation group 1,transplantation group 2,transplantation group 3 and transplantation group 4 became slighter and slighter,while the mice in control group was normal.Average rank's comparisons of the severity of the lung injury between each group had significant difference(P<0.05)except that between bleomycin model groups and transplantation group 1.(4) With the increase of transplanted epithelial progenitor cells to the mice,field planting time in the lung became longer and longer,and there was significant difference between each group(P<0.05).Conclusion Transplantation of epithelial progenitor cells from ESCs may relieve the lung injudy induced by bleomycin,and the action mechanism is associated with cytokines such as TGF-β_1,IFN-γ,etc.
