Study on the relationship between cytokine gene polymorphisms and their serum levels

Abstract: 目的探讨细胞因子基因多态性与其血清水平的关系.方法随机选取65名汉族健康成人,分析其肿瘤坏死因子-α(TNF-α)、转化生长因子-β1(TGF-β1)和白细胞介素-10(IL-10)基因型及分泌水平.细胞因子基因型检测采用序列特异引物聚合酶链反应(PCR-SSP)法,血清细胞因子水平测定采用ABC-ELISA法.结果 TNF-α等位基因型别共有3种,分别为A/A、G/A和G/G,对应的血清TNF-α浓度,A/A组和G/A组均比G/G组高(P<0.01);TGF-β1等位基因型别共有9种,分别为T/T-G/G、T/C-G/G、T/C-G/C、C/C-G/G、T/T-G/C、C/C-G/C、C/C-C/C、T/T-C/C和T/C-C/C,对应的血清TGF-β1浓度,T/T-G/G、T/C-G/G组比T/C-G/C、C/C-G/G和T/T-G/C组高(P<0.01),而T/C-G/C、C/C-G/G和T/T-G/C组又比C/C-G/C、C/C-C/C、T/T-C/C和T/C-C/C组高(P<0.01);IL-10等位基因型别共有6种,分别为GCC/GCC、GCC/ACC、GCC/ATA、ACC/ACC、ACC/ATA和ATA/ATA,对应的血清IL-10浓度,GCC/GCC组比GCC/ACC和GCC/ATA组高(P<0.01),而GCC/ACC和GCC/ATA组比ACC/ACC、ACC/ATA和ATA/ATA组高(P<0.01).结论在受测人群中,细胞因子基因分泌型是血清细胞因子水平高低的决定性因素之一.
