Donepezil protects human umbilical vein endothelial cells against cell injury

Abstract: Objective In this study we investigated the effects of donepezil on the H2O2-induced cell injury of human umbilical vein endothelial cells (HUVEC) and the mechanism of its protective effects.Methods Human umbilical vein endothelial cells (HUVEC) were treated with hydrogen peroxide (H2O2) ; the viability of the cells was then determined using Cell Counting Kit-8 (CCK-8).Cell apoptosis was observed by Hoechst33258 staining.The expression of manganese superoxide dismutase (MnSOD) protein in HUVEC was detected by Western Blot.Results CCK-8 assay showed that donepezil (1 μM) protected the cells from H2O2-induced cell injury significantly (P < 0.05).After Hoechst 33258 staining,the number of cells with apoptotic morphology in donepezil-pretreated group decreased significantly(P< 0.05).Compared with H2O2 group,the expression of MnSOD in HUVECs in donepezil pretreated group was increased.Conclusion Donepezil can protect HUVECs against H2O2-induced cell injury and increase the expression of MnSOD in HUVECs.
