Abstract: Objective To investigate the expression of heat shock protein 70 (HSP70) induced by Geranylgeranylacetone(GGA)and synergistic effect on resistance to Cisplatin in SPCA-I cell line. Methods GGA-induced group and the control group were enrolled. The expression of HSP70 at protein and mRNA levels was analyzed respectively by immunofuorescence and reverse transcription polvmerase chain reaction (RT-PCR). Flow cvtometry was used to determine the effect of Cisplatin on cell apoptosis. Results Compared to the control group, HSP70 expression in both protein and mRNA levels in the GGA-treated group increased. The resistance rates to cisplatin in the control group was 9.70 ±0.63; in the 10, 50, 100,500, 1000 μmol/L GGA-induced groups it was(7.97 ±0. 14)%,(7.78 ± 0.69) %, (6.37 ± 0.15) %, (6.63 ± 0. 93) %, (4.22 ± 0.06) % (P < 0. 05). Flow cytometry assay showed that compared with the control group, GGA-treated cells dramatically decreased. Conclusions GGA affects on the expression of HSP70 in SPCA-I cell line. Up-regulation of HSP70 can signifcantly increase the sensitivity of Cisplatin.
