The experimental study on the effect of alcohol on cardiac tissue in rats

Abstract: Objective To observe the influence of different doses of alcohol on rat cardiac tissue, to establish a chronic animal model and to explore the prevention and treatment of alcoholic heart muscle disease. Methods Totally 140 SD rats were randomly divided into 7 groups: the normal control group and the different doses lavage alcohol model group. Four months later, Jugular Venous blood was immediately taken from each rat to detect creatine kinase isozymes (CK-MB) , cardiac troponin T (cTnT). The myocardial tissue in vivo from rat was taken for pathological examination to observe the impact of alcohol on them. The expression of apoptosis-related protein product Caspase-3 in the myocardial cells was detected. Results The CK-MB, cTnT of A, B groups had no significant change, but CK-MB, cTnT levels in C, D, E, F groups were significantly increased (P<0.01). Myocardial edema, myocardial cell degeneration and necrosis in model D, E, F groups had obvious changes, but the myocardial pathological in model A, B, C groups and the control group had significant changes. Caspase-3 expression rates of model of A, B, C groups had no significant differences, but Caspase-3 expression rate in model of D, E, F groups was obviously higher than that in A, B, C groups and control group (P < 0.05). Conclusions Low dose of alcohol does not cause the increase of cardiac markers and the change of cytopathology. However, excessive alcohol consumption can lead to the increase of myocardial enzymes and Caspase-3 expression, furthermore, the myocardial cell apoptosis or necrosis.
