Abstract: Objective To test if thyroid-stimulating hormone(TSH) can suppress expression of glucose transporter 4(GLUT4)in 3T3-L1 adipocytes. Methods 3T3-L1 preadipocytes were induced to differentiate into adipocytes. These adipocytes were treated with 0 to 1.0 IU/L TSH for up to 48 hours. Protein levels of total GLUT4 were quantified by western blotting. Culture medium interleukin-6 level was assessed by enzyme-linked immunosor-bent assay(ELI(S)A). Results The GLUT4 protein level after 48 hours induced with different dose(0. 01 IU/L, 0.1 IU/L, 1 IU/L) of TSH presented a significant 23% , 44% , 74% ( all P < 0.05 ) reduction compared with control group. While the interleukin-6 level of culture medium increased significantly [( 101. 9300 ± 6. 8396), (104.1533 ±5.9619), (129.0167 ±6.4232) μg/L vs (93.2500 ±2.6695)μg/L, P<0.05]. Conclusion In 3T3-L1 adipocytes, TSH can decrease expression of GLUT43.
