Phosphatidylinositol 3-kinase inhibitor reduces respiratory tract inflammation in a murine model of Asthma

Abstract: Objective To investigate the effect of phosphoinostitide 3-kinase inhibitor on bronchial inflammation in a murine model of asthma. Methods Thirty BALB/c mice were randomly divided into 3 groups (10each group) : PI3K inhibitor (LY294002)-treated group, ovalbumin (OVA) group and normal control group.BALB/c mice were sensitized and challenged with OVA. Cells of bronchoalveolar larvage fluid (BALF) and bone marrow were counted and classified 2 days post last OVA sensitized. Lung tissues were sectioned to hematoxylin-eosin and periodic acid schiff staining. Results LY294002 treatment reduced OVA-induced airway infiltration of inflammatory cells, eosinophils in BALF, inflammatory infiltration and eosinophils numbers around airway, blood vessels or in lung histological sections. Conclusion PI3K inhibitor is effective in treating OVA-induced asthma by attenuating airway inflammation.
