Development of the acute pancreatitis model of rat and effects of hyperbaric oxygen therapy with different pressures

Abstract: Objective To establish an acute pancreatitis model of rat and investigate effects of hyperbaric oxygen (HBO) therapy with different pressures on pancreatitis. Methods Forty SD rats were randomly divided into 3 groups: the sham group ( n = 8 ), the control group ( n = 8 ) that did not receive any treatment and the HBO therapy group ( n = 24 ), which were further divided into 3 subgroups, each consisting of 8 rats, exposed to different pressures. The acute pancreatitis rat model was developed with pancreatic duct ligation. Then, the animals were exposed to HBO with different pressures, and were sacrificed on day 7. Blood and pancreatic tissue samples were collected for analyses. The animals in the sham group were only given celiotomy incision and suture. Pathologic changes in pancreatic tissues were observed under the optic microscope and the transmission electron microscope. Concentrations of inflammatory cytokines (IL-2, IL-6,IL-10, TNF-α) were measured with ELISA. Results Amylase level in the sham group after ligation was (798. 5 ± 113.5 ) IU/L and that in the control group was (3156.2 ± 639.9 ) IU/L. Significant statistical differences could be seen between the 2 groups. Seven days after HBO therapy with different pressures,significant differences in the concentrations of inflammatory cytokines (IL-2, IL-6, IL-10, TNF-α) for the HBO therapy groups and the control group could be noted ( P<0.05 ).And statistical differences in the concentrations of IL-2, IL-6, IL-10, were shown in all the HBO therapy groups(P <0.05). There were statistical differences in the concentrations of TNF-α between the 0.15-0.20 MPa group and 0.15-0.25 MPa group(P <0.05). And no significant differences could be seen between the 0.20 MPa and 0.25 MPa groups (P > 0. 05 ). Pancreatic pathology and electron microscopy indicated that most significant improvement was noted at 0. 25 MPa therapeutic pressure. Conclusions HBO therapy with a pressure of 0. 15- 0. 20 MPa or 0. 25 MPa could significantly alleviate necrosis and hemorrhage of pancreatic glands and inhibit the activity of inflammatory factors in rats with acute pancreatitis.
