The effect of recombinant human growth hormone on the expression of tumor necrosis factor in the rat model of obstructive jaundice

Abstract: 目的 建立梗阻性黄疸(OJ)大鼠动物模型,探讨生长激素(rhGH)对内毒素血症及肿瘤坏死因子α(TNF-α)水平的影响.方法 成年雄性大鼠60只,随机分为假手术(SO)组、梗阻性黄疸(OJ)组、OJ +rhGH组,每组20只.SO组不结扎胆管,OJ组、OJ+rhGH组结扎胆总管.手术2周后每日OJ+rhGH组皮下注射Saizen 0.75IU/Kg,SO组、OJ组相同部位注射等量生理盐水.持续用药两周.实验4周后行内毒素(ETX)、TNF-α、肝功能检测.结果 OJ组血中ETX(0.065±0.011)EU/m1及TNF-α (795.08±350.49)pg/m1水平明显增高,并且两者呈正相关(r=0.847,P<0.01);rhGH+OJ组血中ETX、TNF-α水平分别为(0.036±0.010)EU/ml和(182.00±88.37)pg/ml,与对照组比较差异有统计学意义.结论 rhGH通过降低OJ时ETX,有效降低了其产物脂多糖介导的TNF-α水平.
