All-trans-retinoic Acid Decreases Smooth Muscle Cell Proliferation and Increases Apoptosis in an Animal Model of Vein Bypass Grafting

Abstract: 目的研究全反式视黄酸(atRA)对大鼠自体移植静脉内膜平滑肌细胞(VSMC)增生与凋亡的影响.方法建立40只大鼠自体静脉移植模型,随机分成atRA组和对照组,每组按7、14d随机分成2个亚组,每只大鼠分别于术前4d至术后10d进行atRA(10mg*kg-1*d-1 )或相同剂量的溶剂(植物油)喂养,规定时间点取材固定,Masson染色,免疫组化测定增殖细胞核抗原 (PCNA)及TUNEL法检测平滑肌细胞凋亡,计算机图象分析仪测量移植血管内膜厚度、计算内膜增生指数及细胞增殖和凋亡百分比.结果术后7或14datRA组移植静脉段的内膜厚度、增生指数及 VSMC增殖指数均明显低于对照组(P<0.05).而凋亡指数则明显高于对照组( P<0.05).结论全反式视黄酸(atRA)可以抑制自体移植静脉内膜增生及平滑肌细胞增生,促进平滑肌细胞凋亡.
