Abstract: Objective To explore the relationship between two functional polymorphisms in exons of the PPARγ gene and the susceptibility to T2DM. MethodsPCRRFLP was performed to test the genotypes in 296 newlydiagnosed T2DM cases and 477 controls matched for age and sex. Results The rs3856806 polymorphism of PPARγ gene showed a significant difference between T2DM cases and controls. Compared with CC genotype, those carrying CT/TT genotype had a lower T2DM risk. In the stratified analysis, compared with CC genotype, those carrying CT/TT genotype had a significantly reduced risk in female,the elders (aged above 50 years),hypertensive subjects and normal weight individuals with OR value of 0.33(95%CI: 0.14, 0.76), 0.45(95%CI: 0.23, 0.88), 059(95%CI: 0.36, 0.95) and 0.34(95%CI: 0.13, 0.94), respectively. There was no significant difference in genotype frequency of rs1801282 polymorphism between T2DM patients and controls. However, the obese individuals carrying CG(Pro/Ala) genotypes had a lower T2DM risk. Conclusions The polymorphisms of PPARγ gene might play a role in the susceptibility of T2DM in Han population of Jiangsu.
