Abstract: Objective To investigate the association of clusterin (Clu) gene polymorphism with type 2 diabetes mellitus (T2DM) Methods Clu gene was screened by PCR-DGGE in 62 cases with type 2 diabetes mellitus,60 individuals as normal control who were without history of cardiovascular,hepatic and renal disorders.Genomic DNA were amplified using GC-clamped primers covering Clu exons 2 and 5 regions.Abnormal bands DNA were sequenced. Results 1963 (+A) polymorphism in exon 2 of the Clu gene was detected. Three novel genetic polymorphisms were found in exon 5 of Clu:①7476 (-A),②7534 (-C),③7521 (C→T). Conclusions These polymorphisms are significantly different between the normal control group and the type 2 diabetes mellitus group.These polymorphisms may be associated with type 2 diabetes mellitus
