Abstract: 目的 构建人过氧化物酶体增殖体激活受体γ2 (human peroxisome proliferator activated receptor γ2,hPPARγ2)真核表达载体并建立稳定表达hPPARγ2的细胞株.方法 用RT-PCR扩增hPPARγ2的全长cDNA并构建出pcDNA3.1(+)/PPARγ2真核表达载体,经核苷酸序列分析证实.真核表达载体pcDNA3.1(+)/ hPPARγ2转染到HESF细胞,G418筛选,获得稳定表达hPPARγ2的细胞株,检测其在HESF细胞中的表达.结果 核苷酸序列测定证实获得hPPARγ2真核表达载体,RT-PCR、间接免疫荧光染色和Western-blot结果显示转染hPPARγ2的细胞株稳定表达hPPARγ2.结论 构建了hPPARγ2真核表达载体,并获得稳定表达hPPARγ2的细胞株,为进一步研究hPPARγ2的功能奠定基础.
