Effect of advanced glycation end products on activity and expression of platelet endothelial nitric oxide synthase in the normals

Abstract: Objective To probe the mechanism of damage of platelet function by observing the effect of advanced glycation end products (AGEs) on both activity and expression of platelet endothelial nitric oxide synthase(eNOS) in the normals. Methods Peripheral venous blood was collected from six normal subjects. Platelets isolated with gel-filtration chromatography were incubated with three concentrations of AGEs(100 μg/ml, 200 μg/ml, 400 μg/ml)for 30 minutes. And then platelets were divided into two aliquots: one used for NOS activity measurement, which was determined by 3H L citrulline formation from 3H L arginine;and the other for purifying eNOS with immunoprecipitation method. eNOS expression was detected by western blot. Results AGEs significantly inhibited platelet NOS activity of the normals in a concentration dependent manner. eNOS was highly expressed in platelets; the expression of platelet eNOS had no significant reduction after platelets were incubated with AGEs for 30 minutes. Conclusion The inhibition of platelet eNOS activity by AGEs facilitates platelet activation, which had no relation to eNOS expression.
