Effect of epinephrine and glucose on aquaporin adipose mRNA expression in 3T3-L1 Adipocytes

Abstract: Objective Aquaporin adipose(AQPap) is the physiological glycerol channel specific to adipocytes, and it is expressed predominantly in adipose tissue. In this study, we investigated the effect of epinephrine and glucose on expression of AQPap in 3T3 L1 adipocytes, in order to know what role AQPap plays in obesity and diabetes mellitus. Methods The differentiated 3T3 L1 adipocytes were incubated with 10 -6 mol/L?10 -7 mol/L?10 -8 mol/L?10 -9 mol/L epinephrine for 6 hour, or stimulated with different levels of glucose for 48 hours, then total cellular RNA were extracted, and the expression of AQPap mRNA were detected by RT PCR. Results There was no significant change in AQPap mRNA levels under the treatment with various concentrations of epinephrine ( P >0.05).After stimulation with high concentration of glucose, AQPap mRNA expression was augmented in 3T3 L1 cells ( P <0.05) With stimulation of 16.8 mmol/L glucose, the AQPap mRNA level was three times increased, compared with the control(glucose concentration was 5.6 mmol/L ). Conclusion Epinephrine, one of the lipolytic hormones, has no effect on the expression of AQPap mRNA levels, but high level glucose can increase AQPap mRNA expression.
