Effect of phenolic acids isolated from Sambucus williamsii on proliferation and differentiation of rat osteoblastic UMR106 cells

Abstract: 目的研究接骨木Sambucus williamsii Hance中抗骨质疏松的活性成分,及其对UMR106类成骨细胞增殖和分化的影响.方法以大鼠类成骨细胞UMR106的增殖为指导活性成分分离的指标,利用各种分离手段追踪分离接骨木茎枝60%乙醇提取物活性部位的化学成分,并利用光谱学方法鉴定其结构,检测对UMR106细胞增殖和分化的影响.结果从接骨木中分离得到了7个酚酸类化合物,分别鉴定为香草醛(vanillin,Ⅰ)、香草乙酮(acetovanillone,Ⅱ)、松柏醛(coniferyl aldehyde,Ⅲ)、丁香醛(syringaldehyde,Ⅳ)、对羟基苯甲酸(4-hydroxybenzoic acid,Ⅴ)、对羟基桂皮酸(4-hydroxycinnamic acid,Ⅵ)、原儿茶酸(protocatechuic acid,Ⅶ).检测了化合物Ⅰ、Ⅲ~Ⅴ和Ⅶ对UMR106细胞活性的影响.结论7个酚酸类化合物均为首次从接骨木中分离得到,化合物Ⅳ可同时促进UMR106细胞的增殖和分化,化合物Ⅰ和Ⅲ可促进细胞的增殖,化合物Ⅴ和Ⅶ可促进细胞碱性磷酸酶活性.
