Abstract: Objective To screen specific proteins in patients with hypertrophic scar via comparing the distinction in proteomics to normal skin.Methods Hypertrophic scar tissues (8 patients with hypertrophic scar)and normal cutaneous tissues (3 patients underwent trunk plastic surgery) were collected from Department of Burns Surgery,The First Affiliated Hospital of Sun Yat-sen University,between January 2010 and May 2010.The distinction in proteomics between hypertrophic scars and normal cutaneous tissues was compared via twodimensional gel electrophoresis.The protein spots with differential expression were subjected to MALDI-TOF/TOF mass spectrometry and bio-information analysis.Results Two-dimensional electrophoresis fluorescent staining atlas with high repeatability was obtained,showing an average of 2975 protein spots in hypertrophic scar and 3053 in normal cutaneous tissue.Further analyses on the spots with a minimum of 4-fold distinction identified 24proteins ( 11 up-regulated and 13 down-regulated) via mass spectrometry and database searching.Conclusion Successful establishment of two-dimensional electrophoresis fluorescent staining atlas is helpful to identification of differential proteins between hypertrophic scar and normal skin.
