Identification of a positively 5' regulatory region in human Toll like receptor 4 gene

Abstract： 目的 初步确定Toll样受体4(Toll like receptor 4,TLR4)基因5'远端髓系细胞特异性的转录调控区域.方法 采用分子克隆结合报告基因分析的方法,构建TLR4基因5'旁侧-2413～+66序列与荧光素酶报告基因载体pGL-3的重组质粒及其缺失质粒,通过DEAE-Dextran介导的转染技术导入人K562细胞,检测各报告基因质粒荧光素酶活性.结果 TLR4基因5'旁侧转录起始位点上游-1337～-683 bp区域在K562细胞中,能够增强报告基因转录活性.结论 TLR4基因5'旁侧存在远端细胞特异性的调控区域,定位并鉴定参与调控的转录因子,可望阐明TLR4基因表达调控及其组织和细胞特异性表达的机制.
