Effects of ketamine on neuron apoptosis and expression of Caspase-3 and c-fos in gerbils with cerebral ischemia/reperfusion injury

Abstract: 目的研究氯胺酮对沙土鼠脑缺血-再灌流损伤后细胞凋亡状态及相关基因caspase-3、c-fos表达的影响.方法沙土鼠随机分为假手术组、缺血-再灌流组和氯胺酮组,各10例.缺血-再灌流组:分离双侧颈总动脉,用无创动脉夹阻断脑血流10min后松开动脉夹恢复血流.假手术组:仅分离双侧颈总动脉但不予阻断.氯胺酮组:血流阻断前30min腹腔内注射氯胺酮10mg/kg,余同缺血-再灌流组.采用缺口末端标记法(TUNEL)原位标记DNA片段检测凋亡细胞,免疫组化SP法进行caspase-3、c-fos的免疫组织化学染色.结果缺血-再灌流组及氯胺酮组脑组织中TUNEL及caspase-3、c-fos阳性细胞数明显多于假手术组(P<0.01);氯胺酮组3项指标阳性细胞数目均明显低于缺血-再灌流组(P<0.01).结论氯胺酮减少沙土鼠脑缺血-再灌流损伤后细胞的凋亡,其对caspase-3激活和c-fos表达的抑制可能为此作用的机制之一.
