Influences of nitric oxide and leukocytes infiltration on acute focal cerebral ischemic reperfusion in rats

Abstract: 目的观察一氧化氮(NO)和白细胞浸润对急性局灶性脑缺血再灌流损伤的作用.方法建立大鼠急性大脑中动脉栓塞(MCAO)及再灌流模型,采用NADPH-d 组织化学方法检测大脑皮质神经元内NOS活性,利用MPO活性测定白细胞浸润.结果①在脑缺血15min、30min再灌流4h时,NOS、MPO活性均随着脑缺血时间延长而增加(P<0.01);②在脑缺血60min再灌流4h时,NOS活性明显降低(P<0.05),MPO活性显著增加(P<0.05).结论实验结果说明NO与白细胞浸润在急性脑缺血早期共同作用脑缺血-再灌流损伤;在急性脑缺血-再灌流后期,脑缺血再灌流损伤主要与白细胞浸润有关.
