Abstract: 目的 探讨脱氢穿心莲内酯琥珀酸半酯(DAS)通过抑制iNOS减轻LPS诱导的急性肺损伤导致的氧化应激.方法 30只雄性BALB/C小鼠平均分为对照组(Control组)、治疗组(LPS+DAS组)和模型组(LPS组).使用ELISA对肺泡灌洗液(BALF)中IL-1β、IL-6和TNF-α水平进行测定.测量BALF中丙二醛(MDA)和超氧化物歧化酶(SOD)水平.对肺组织进行湿/干比(wet to dry ratio,W/D)测定.HE染色用于肺组织病理变化观察和肺组织损伤评分测量.RT-PCR被用于iNOS mRNA的测定.使用Western blot测定iNOS和β-肌动蛋白(β-actin)蛋白的表达.结果 LPS干预后小鼠BALF中IL-1β、IL-6、TNF-α和MDA水平明显上升而SOD水平显著下降,W/D比值和肺组织损伤评分明显升高,iNOS mRNA和蛋白的表达明显增加.LPS+DAS组小鼠BALF中IL-1β、IL-6、TNF-α和MDA水平、W/D比值、肺组织损伤评分、iNOS mRNA和蛋白的表达较LPS组小鼠明显下降;同时BALF中SOD水平较LPS组小鼠明显增加.结论 DAS可能是通过抑制iNOS表达减轻LPS诱导的急性肺损伤导致的氧化应激.
