Abstract: AIM:To study the effect and mechanism of Janus Kinase2-selective inhibitor AG490 in proliferation,apoptosis and the capacity of invasion and metastasis of human cervical cancer Hela cells.METHODS:The cervical cancer cell line Hela cell was treated with AG490 in different doses.MTT assay,flow cytometry and Western Blot were performed to detect the state of cell proliferation,the cell cycle and apoptosis,proteins expression of Stat3,P-Stat3,Mmp-2,Vegf,respectively.RESULTS:AG490 could remarkably inhibit the proliferation of cervical cancer cells,block the cell cycle,facilitate the apoptosis of cervical cancer,and result in less expression of P-Stat3,Mmp-2,Vegf.CONCLUSION:Blocking JAK2/STAT3 signaling pathway activation with AG490 can inhibit the proliferation and promote apoptosis ability of cervical cancer ceils,down-regulate protein expression of P-Stat3,Mmp-2 and Vesf,inhibit the capacity of invasion and metastasis of cervieal cancer Hela cell.Blocking JAKs/STATs signaling pathway may become a novel strategy in treatment and prevention of invasion and metastasis of cervical cancer.
