Astragalus membranaceu reduces ischemia-reperfusion injury of rat liver graft by inhibiting NF-κB

Abstract: 目的:探讨黄芪(AM)对大鼠移植肝脏缺血再灌注损伤的保护作用及机制,方法:选用128只雄性健康SD大鼠,随机分成黄芪给药组(AM组)和生理盐水对照组(NS组),以二袖套法建立原位肝移植模型.术后1,2,4 h检测血清丙氨酸转氨酶(ALT)值,ELISA检测肿瘸坏死因子-α(TNF-α)值,Western Blot检测肝组织细胞间黏附分子-1(ICAM-1)的表达,免疫组化法及ELISA测定肝组织核因子-κB(NF-κB)的表达,术后3 d取肝脏标本做HE染色检查,TUNEL法检测肝脏细胞凋亡.结果:在肝移植完成后AM组ALT分别为(9.4±0.4),(10.8±0.5),(14.3±1.1)μkat/L,较NS组的(12.4±0.6),(15.9±0.8).(18.7±1.1)μkat/L低(P<0.05);AM组TNF-α水平分别为(757±54),(977±60),(318±53)ng/L,较Ns组(1258±132),(1831±165),(499±66)ng/L低(P<0.05);AM组的NF-κB,ICAM-1水平在各个时间点均低于NS组(P<0.05);AM组肝细胞凋亡程度低于NS组(P<0.05);AM组肝脏组织的病理改变较NS组肝脏组织的损伤轻.结论:黄芪能减少炎症介质释放,减轻移植后肝脏结构和功能上的破坏,提高肝脏耐受缺血再灌注损伤的能力.
