Impact of gshb gene mutant of Pseudomonas aeruginosa on expression of MexXY-OprM effiux pump

Abstract： 目的:研究铜绿假单胞菌谷胱甘肽合成酶基因(gshb)突变体对MexXY-OprM外排泵表达的影响.方法:运用同源重组技术构建铜绿假单胞菌的谷胱甘肽合成酶基因(gshb)突变体,将连有MexXY-OprM外排泵启动子的pMS402质粒电转化到此突变体中,通过pMS402质粒上的Lux报道子发光值大小检测MeXY-OprM外排泵的表达变化情况.结果:MexXY-OprM外排泵的表达在谷胱什肽合成酶突变体中显著降低.结论:铜绿假单胞菌中的谷胱甘肽对MexXY-OprM的表达具有促进作用,这将为进一步探讨MexXY-oprM的分子机制提供理论依据.
