Abstract: 目的:研究人DeltaNp73α基因重组腺病毒转染人脐血来源树突状细胞(DC)对其成熟状态及其凋亡水平的影响.方法:用Adeasy系统构建人DeltaNp73α基因重组腺病毒;并通过增强离心法将其转染至人脐血来源DC;流式细胞仪分析转染后DC的成熟状态以及DC的凋亡水平.结果:成功构建了人DeltaNp73α基因重组腺病毒并高效转染DC;人DeltaNp73α基因重组腺病毒转染提高了DCCD83,HLA-DR的表达,并能显著抑制其凋亡水平.结论:人DeltaNp73α基因重组腺病毒转染DC,促进DC的成熟并抑制其凋亡,将有效地提高DC疫苗的抗原呈递能力.
