Abstract: 目的:探讨反义c-myc寡核苷酸(c-myc ASODN)对半乳糖诱导的兔晶状体上皮细胞(LEC)中c-Myc蛋白表达及细胞周期的影响.方法:人工合成与c-myc基因第二外显子翻译起始区序列互补的寡核苷酸并磷酸化修饰,用半乳糖和c-myc ASODN处理体外培养的兔晶状体,流式细胞术检测c-myc ASODN对半乳糖诱导的LEC中c-Myc蛋白表达水平及细胞周期的影响.结果:2.5~15.0 μmol/L c-myc ASODN能下调半乳糖诱导的晶状体上皮细胞中c-Myc蛋白的表达,荧光强度从214.7±0.6降至90.5±8.4,其作用有剂量依赖性(r=-0.983,P<0.01),同时可降低S期细胞数,从5.54降至4.59.结论:c-myc ASODN能抑制半乳糖诱导的晶体上皮细胞中c-Myc蛋白的表达水平;阻止细胞进入S期,抑制细胞增殖.
