Abstract: 目的比较两种介导物多阳离子脂质体(lipofectamine)及一种具有专利的脂类混合物(FuGENE6)的基因转移效率. 方法体外培养人视网膜色素上皮细胞(RPE),并以携带β-半乳糖苷酶报道基因的多阳离子脂质体和脂类混合物转移人视网膜色素上皮细胞,以X-gal染色系统检测. 结果分别以多阳离子脂质体及脂类混合物转移6 h和表达48 h后, 在人视网膜色素上皮细胞中β-半乳糖苷酶的阳性表达比例为47%和6%. 结论在体外能够由第二代多阳离子脂质体有效地将外源基因转移至人视网膜色素上皮细胞 ,脂类混合物仅获得较低的转染效率.
