Construction of phage display library of IgG1 Fab antibodies from lymph nodes of gastric cancer patients

Abstract： AIM To construct a phage display library of hu-man IgG1 Fab antibodies from lymph nodes of gastric cancer patients using phage display technique. METHODS Total RNA was isolated from lymph nodes of gastric cancer patients. Fd fragment and kappa light-chain were amplified by RT-PCR and cloned into the phagemid pCOMB3 and expressed as a fusion protein with phage M13 PⅢ in E.coli XL1-Blue.In the presence of help phage M13K07, the Fab fusion protein was displayed on the surface of recombinant phage. RESULTSWe have constructed a phage display library of human IgG1 Fab antibodies whose titer was about 2.56×1015 pfu*L-1. CONCLUSION This library could be used to prepare human IgG1 Fab antibody.
